Design Notes
The TEV recognition sequence encoded in the rough center of this part is flanked on both sides by sequences that code for glycine-serine-glycine linkers. This feature was designed to serve as a homologous region allowing for facile substitution of either a receptor or a transactivator around the TEV recognition sequence using the Gibson ligation assembly method.
Source
This part includes human histamine receptor 4 (HRH4), the tobacco etch virus (TEV) protease recognition sequence, the DNA-binding domain from the Gal4 transactivator of Saccharomyces cerevisiae, and the VP16 transactivation domain from the herpes simplex virus.
